RESUMO

Sea cucumbers are one of the most common large animals in the deep-sea benthic communities, and those of the genus Pannychia are particularly abundant in the bathyal North Pacific Ocean. In Japanese waters, three species of Pannychia have been reported, mainly from the northern and eastern parts of the country. Here, we describe two new species from the western part of Japan. These two new species were placed in Pannychia by the presence of dorsal papillae on the ventrolateral radii and the presence of large wheel ossicles with rim teeth. They are distinguished from all other nominal species of Pannychia on the basis of the morphological characteristics of their body wheel ossicles. Pannychia nagasakimaruae sp. nov. has a unique ossicle composition in its dorsal and ventral body walls: four forms of wheel ossicles. Pannychia rinkaimaruae sp. nov. differs from other congeners in its small wheel ossicles in the dorsal and ventral body walls, which have conical rim teeth. We barcoded partial COI gene sequences from holotypes and paratypes of two new Japanese species. Our molecular phylogenetic analysis showed that these two new morphologically recognized Japanese species form distinct clades separated from other Pacific Pannychia species, so that the two new Japanese species were also supported by genetic results. In addition, we describe and visualize the poorly calcified calcareous rings of the two new species using X-ray µCT system, a useful method for observing such structures, which are important structures in holothurian anatomy.

RESUMO

The phyllophorids Phyllophorella robusta, Selenkiella siamense and Cladolella virgo, as well as the sclerodactylid Clarkiella discoveryi, are the type species of their respective genera, all described by Heding Panning (1954) and each known only from a single specimen, were revisited in order to reassess their morphology by means of optical and electron microscopy, and micro-computed tomography. Additionally, a new species of phyllophorid is described, the fourth of the genus Thorsonia.

RESUMO

A new species of Dendrochirotida sea cucumber, Parathyone itapuaensis, is described and illustrated herein based on specimens collected from rocky intertidal regions of the northeastern coast of Brazil. The new species is characterized by a suite of morphological characters revealed by light stereomicroscope, SEM and microCT studies of the calcareous ring, dermal ossicles, Polian vesicles and madreporite. Parathyone itapuaensis sp. nov. is characterized by having 10 tentacles, two Polian vesicles and single madreporite; length of anterior process (radial plate) larger than length of the base of the radial plate; body wall with buttons with triangular knobs and knobbed multiperforated plates in the anal region; and tentacles with arched rods, knobbed in the margins and shallow cups knobbed in the outer and inner surfaces. A key to the recognition of Parathyone species is given.

Introduction: The exploitation of the sea cucumber (Holothuria (Panningothuria) forskali) in Ría de Vigo (NW Spain) is recent (2015) and it has been done until now with scarce or no information about some key biological aspects as reproduction, recruitment or growth. Objective: To describe the reproductive cycle of H. forskali in Ría de Vigo. Methods: We sampled fortnightly throughout 2018. We calculated gonadal condition indices (GCI) and gametogenic stages by classic histological methods. Results: The reproductive cycle of H. forskali in the Ría de Vigo is characterized by a sexual resting stage during spring, when temperature and daylight hours are lower; the beginning of gametogenesis during summer, when temperature is higher, daylight hours longer and the sea bottom is rich in nutrients; then, a period of spawns interspersed with a gonadal restoration during autumn and winter, when temperature is lower and food is scarce. Sex ratio is 1:1, however, the studied population is not synchronized, because females initiate maturation earlier. The comparison of the histological results with GCI indices suggest that GCI is a good indicator for gonadal stage. Fishery management strategies, such as a closure period, must be adapted to the reproductive stage. We recommend avoiding fishing between November and February to increase spawning potential ratio and, consequently, recruitment.


Introducción: La explotación del pepino de mar (Holothuria (Panningothuria) forskali) en la Ría de Vigo (NO España) es reciente (2015) y se ha llevado a cabo con poca o ninguna información sobre algunos aspectos biológicos claves como la reproducción, el reclutamiento o el crecimiento. Objetivo: Describir el ciclo reproductivo de H. forskali en un banco de la Ría de Vigo. Métodos: Se tomaron muestras quincenales durante el 2018. Se calcularon índices de condición gonadal (ICG) y se obtuvieron los estados gonadales siguiendo métodos histológicos clásicos. El estado gonadal de cada individuo se asignó de acuerdo con una escala modificada por otros autores, y en ella se describe un nuevo estado de recuperación gonadal, no descrito hasta ahora para esta especie. Resultados: El ciclo reproductivo de H. forskali se caracteriza por una etapa de reposo sexual en primavera, un inicio de la gametogénesis durante el verano y un período de puestas sucesivas intercaladas con recuperaciones gonadales durante el otoño e invierno. La proporción de sexos es 1:1 y la población estudiada no está sincronizada, las hembras inician la maduración antes. La comparación de los resultados histológicos con los índices ICG sugiere que este índice es un buen indicador del estado gonadal. El período de veda debe adaptarse a la etapa reproductiva, por lo que recomendamos evitar la extracción del recurso al menos entre noviembre y febrero para asegurar un mayor número de puestas y, en consecuencia, el reclutamiento.

RESUMO

Functional studies on echinoderms have been reduced to the use of pharmacological treatments. The ability to modulate the genetic expression of regenerating tissues can elucidate potential effectors during this process. Here we describe an effective transfection protocol that allows the introduction of Dicer-substrate interference RNAs (DsiRNAs) for the modulation of gene expression and its characterization during regeneration.

The antiviral effect against RVA in cell culture was evaluated by using an aqueous extract of Patallus mollis sea cucumber, applying the titration methodology. This technique is used to measures the ability of the extract dilutions to inhibit the cytopathic effect (CPE) of the virus, expressed as percentage of inhibition (IP). The mean extract cytotoxic concentration (CC50) used in the antiviral assay was 27,042.10 µg/mL and the PI of the antiviral activity extract was greater than 99.9% for each concentration. To determine the viral action mode, the cells were previously treated with the extracts in different stages during the viral infection cycle. The result analysis suggests that the extract inhibits 99% of the virus during the absorption and viral inactivation phase. These results show the P. mollis extract has a remarkable antiviral effect against the RVA in cell culture. So that, it is crucial to investigate its action mechanisms.

Despite descending from heavily calcified ancestors, the holothuroid skeleton is fully internal and composed of microscopic ossicles and a ring of plates bound by connective tissue, the calcareous ring. The calcareous ring exhibits a complex and poorly understood morphology; as a result, establishing unambiguous homology statements about its macrostructure has been challenging and phylogenetic studies have had to simplify this important structure. Here, we provide the first broad comparative study of Dendrochirotida calcareous rings using micro-computed tomography (µCT). A detailed description of the three-dimensional macrostructure of the calcareous ring of 10 sea cucumber species, including rare and type specimens, is presented. The structures observed were highly variable at the subfamily level, especially at the point of tissue attachment. The relationship between the calcareous ring and its associated organs, and their functional morphology are discussed. To aid future phylogenetic studies, we listed 22 characters and performed a preliminary cladistic analysis. The topology obtained supports the idea that the simple, cucumariid ring is ancestral to the mosaic-like phyllophorid ring; however, it did not support the monophyly of the cucumariids. It also did not support the family Sclerodactylidae, which was described based on the ring morphology. Differently from the dermal ossicles, which are highly homoplastic, the general homoplasy index of the calcareous ring characters was relatively low. This result highlights the importance of this structure for phylogenetic inference. Unfortunately, time since collection, rough collection methods and fixation can damage the skeleton, and the calcareous ring is often overlooked in taxonomic descriptions. The data presented here will improve our understanding of holothuroid relationships and facilitate studies on holothuroid functional morphology and biomechanics.

While age is fundamental in animal biology, forming the basis of critical concepts such as life-history strategies, longevity and population structures, measuring this variable in some taxa remains problematic. Such is the case of holothuroid echinoderms, which play key roles in marine benthic communities from the shore to the abyss, and which are extensively fished in many regions across the globe. Here, we present and validate a promising ageing technique using the cold-water species Psolus fabricii. The method involves the extraction of the oldest dermal plates (largest dorsal ossicles) to preserve their original pigments and structure. While plates initially appear to have a uniform texture, polishing and dying them reveals layered ring patterns. A study of laboratory-reared juveniles, from settlement to 40 months of age, confirmed that one layer is added annually, making plates both larger and thicker, and generating successive light and dark rings, the latter representing the transition (overlap) between two layers. Therefore, each pair of rings represents an annual growth band. Size-at-age data obtained using this method revealed that growth of P. fabricii is slow and that wild individuals collected at diving depths had reached an age of several decades.

Reconstructing the evolutionary assembly of animal body plans is challenging when there are large morphological gaps between extant sister taxa, as in the case of echinozoans (echinoids and holothurians). However, the inclusion of extinct taxa can help bridge these gaps. Here we describe a new species of echinozoan, Sollasina cthulhu, from the Silurian Herefordshire Lagerstätte, UK. Sollasina cthulhu belongs to the ophiocistioids, an extinct group that shares characters with both echinoids and holothurians. Using physical-optical tomography and computer reconstruction, we visualize the internal anatomy of S. cthulhu in three dimensions, revealing inner soft tissues that we interpret as the ring canal, a key part of the water vascular system that was previously unknown in fossil echinozoans. Phylogenetic analyses strongly suggest that Sollasina and other ophiocistioids represent a paraphyletic group of stem holothurians, as previously hypothesized. This allows us to reconstruct the stepwise reduction of the skeleton during the assembly of the holothurian body plan, which may have been controlled by changes in the expression of biomineralization genes.

Apart from sharing common ancestry with chordates, sea cucumbers exhibit a unique morphology and exceptional regenerative capacity. Here we present the complete genome sequence of an economically important sea cucumber, A. japonicus, generated using Illumina and PacBio platforms, to achieve an assembly of approximately 805 Mb (contig N50 of 190 Kb and scaffold N50 of 486 Kb), with 30,350 protein-coding genes and high continuity. We used this resource to explore key genetic mechanisms behind the unique biological characters of sea cucumbers. Phylogenetic and comparative genomic analyses revealed the presence of marker genes associated with notochord and gill slits, suggesting that these chordate features were present in ancestral echinoderms. The unique shape and weak mineralization of the sea cucumber adult body were also preliminarily explained by the contraction of biomineralization genes. Genome, transcriptome, and proteome analyses of organ regrowth after induced evisceration provided insight into the molecular underpinnings of visceral regeneration, including a specific tandem-duplicated prostatic secretory protein of 94 amino acids (PSP94)-like gene family and a significantly expanded fibrinogen-related protein (FREP) gene family. This high-quality genome resource will provide a useful framework for future research into biological processes and evolution in deuterostomes, including remarkable regenerative abilities that could have medical applications. Moreover, the multiomics data will be of prime value for commercial sea cucumber breeding programs.

The regeneration of the anterior portion of the body after fission was studied in the holothurian Cladolabes schmeltzii using electron microscopy methods. Following fission, the posterior portion of the digestive tube, cloaca, and respiratory trees remain in the posterior fragment of the body. The regeneration comprises five stages. In the first stage, connective-tissue thickening (an anlage of the aquapharyngeal bulb) occurs on the anterior end between the torn-off ends of the ambulacra. Most of the lost anterior organs developed in the second and third stages. The structures of water-vascular system and nerve ring form through dedifferentiation, proliferation, and migration of cells of the radial water-vascular canals and the radial nerve cords, correspondently. The lost digestive system portion is restored through the formation and merging of two anlagen. The digestive epithelium of the esophagus and pharynx develops from lining cells of microcavities near the central portion of the connective-tissue thickening, which probably migrate from the epidermis. The second gut anlage develops through transformation of the anterior gut remnant portion. The enterocytes partly dedifferentiate, but the epithelium retains integrity. The gut anlage grows down the mesentery and joins the regenerating aquapharyngeal bulb. In the fourth and fifth stages, all lost organs are formed and have nearly normal structure. The regeneration was concluded to occur through morphallactic rearrangements of the remaining parts of organs. Epithelial morphogenesis is the key development mechanism of the digestive, water-vascular, and nervous systems.

The 2009 expedition with the research vessel Miriky sampled the sublittoral and bathyal waters of the northern Mozambique Channel. This exploration campaign resulted in a small, but very diverse collection of holothuroids comprising 174 specimens representing 31 species, 18 genera, 10 families and 5 orders. Of these species, many were hitherto unknown for Madagascar or even for the Indian Ocean, and six, Bathyplotes aymeric sp. nov., Holothuria (Cystipus) yann sp. nov., Holothuria (Stauropora) bo sp. nov., Holothuria (Metriatyla) alex sp. nov., Holothuria (Theelothuria) cyrielle sp.nov., Molpadia thandari sp. nov., are new to science. Molpadia lenticulum (Cherbonnier & Féral, 1981) is a new combination. This contribution provides an illustrated and annotated overview of the poorly known, highly biodiverse, sublittoral and bathyal sea cucumber fauna of the northern Mozambique Channel. Our findings demonstrate how ignorant we are about the poorly explored habitats of our planet and therefore stress the urgent need for more explorations to such regions.

A new genus Hemiocnus is here erected to accommodate the Mediterranean dendrochirotid sea cucumber Cladodactyla syracusana Grube, currently classified, with some doubt, in the cucumariid genus Pseudocnella. At the same time a new cucumariid species, Hemiocnus rubrobrunneus, is described from some Tunisian material, misidentified as Pseudocnella syracusana (Grube), received from the United States National Museum. The new genus appears most closely related to Pseudocnella than to any other genus within the Colochirinae. Although its body wall ossicles resemble those of Pseudocnella spp. it differs in that the two ventral-most tentacles are reduced and in the presence of rosettes in the tentacles. P. syracusana also cannot be classified in Ocnus because of the presence of multi-layered, fir-cone shaped plates in the body wall, often with one end denticulate; such ossicles are lacking in the type species of the latter genus. The new species, Hemiocnus rubrobrunneus, on the other hand, shows some resemblance to H. syracusanus in its characteristic buttons and incomplete baskets, differing in its softer body wall, lack of fir-cone-shaped plates and in the presence of rosettes and complete baskets in the body wall. There are also some resemblances of the new species to the Mediterranean species of Ocnus viz. O. brunneus, O. planci and O. lacteus, but the soft nature of the body wall, shallow quadrilocular instead of deep trilocular baskets, and the presence of large knobbed plates in the anal region precludes its inclusion in this genus.

Most species of Taeniogyrus Semper, 1867 are known from shallow water in the Indo-Pacific, with other records in Antarctica, Mediterranean Sea, and the Atlantic. A new species of Taeniogyrus is described and illustrated here from the continental slope of Campos Basin, southeast of Brazil. In this species, sigmoid hooks (336-405 µm) are much larger than in any other in the genus, bearing a long and conspicuous hook region. Wheels with six spokes (86-169 µm), inner margin with 60-125 continuous teeth, are confined to round papillae along each interradius. Polian vesicles are ventral, numerous (15-21), of different sizes, and tubular shaped with a terminal round region. This new species represents the deepest record of the genus Taeniogyrus. It increases to three the number of chiridotids in Brazilian waters, and the number of Taeniogyrus species in the Atlantic. Additionally, Taeniogyrus furcipraeditus (Salvini-Plawen, 1972) from the Mediterranean Sea and Taeniogyrus havelockensis (Rao, 1975) from the Andaman Sea are proposed as new combinations.

The case put by Alexei Smirnov in 2012 is accepted and the order name Synaptida Cuénot is adopted in place of Apodida Brandt. Two new Synaptida species are described for the Weddell Sea in Antarctica with single author O'Loughlin: Sigmodota magdarogera sp. nov. and Taeniogyrus bamberi sp. nov.. A specimen of Sigmodota magnibacula (Massin & Hétérier) is described. A key is provided for the genera and species of Taeniogyrinae that occur south of the Antarctic Convergence.

Twenty four specimens of holothuroids recently received from the South African Environmental and Observation Network (SAEON), collected from off the south and west coasts of South Africa, are herein recorded and/or described. The specimens comprise eight nominal and one indeterminate species and represent both shallow-water and deep-sea forms, distributed from Plettenberg Bay to just north of Lambert's Bay in the Western Cape Province. There are no new species but two new records for the South African region and extensions of horizontal and bathymetric distributions of the other species. Additions to the South African fauna are Zygothuria lactea (Théel, 1886) and Synallactes cf. challengeri (Théel, 1886). The paper also contains the first definite record of Thyone venusta Selenka, 1868, originally described from the Red Sea. Distribution ranges of the following species have been altered Synallactes viridilimus Cherbonnier, 1952; S. mollis Cherbonnier, 1952 and Psuedostichopus langeae Thandar, 2009.

Resumen Los equinodermos del Pacífico Sur mexicano han sido estudiados por tres siglos, sin embargo discrepancias en la nomenclatura de algunas especies han prevalecido a través del tiempo. El objetivo de este trabajo es presentar la primera lista de especies actualizada de todos los nombres válidos y sinonimias, así como una revisión histórica del estudio de los equinodermos en el Pacífico Sur mexicano. La lista de especies se basa en una exhaustiva búsqueda de registros de literatura y de especímenes depositados en colecciones de referencia. Existen 162 especies válidas de equinodermos en el Pacífico Sur mexicano incluidas en 96 géneros, 54 familias y 20 órdenes. El estado de Guerrero presentó 135 especies, Oaxaca, 94 y Chiapas, 15. Se actualizó la lista de especies y se añadieron cinco nuevos registros de especies (Microphiopholis platydisca, Ophiostigma tenue, Arbacia stellata, Thyone bidentata, Chiridota rigida) para el Pacífico Sur mexicano y uno para el Pacífico mexicano (Encope laevis). Esta lista de especies incrementa el número de especies para el área de estudio, sin embargo, los estudios sugieren que aún existen más especies por ser descubiertas.


Abstract The echinoderms of the Southern Mexican Pacific have been studied for three centuries, but discrepancies in the nomenclature of some species have pervaded through time. The objective of this work is to present the first updated checklist of all valid species and synonyms, and a historical review of the study of the echinoderms of the Southern Mexican Pacific is also presented. The checklist is based on an exhaustive published literature search and records of specimens deposited in museum and curated reference collections. There are 162 species of echinoderms in the Southern Mexican Pacific from 96 genera, 54 families and 20 orders. The State of Guerrero presented a total of 135 species, Oaxaca 94 and Chiapas 15. We updated the list and added five new records of the species (Microphiopholis platydisca, Ophiostigma tenue, Arbacia stellata, Thyone bidentata, Chiridota rigida) for the Southern Mexican Pacific and one for the Mexican Pacific (Encope laevis). This checklist expands the number of species known for the study area; nevertheless studies suggest that still more species are yet to be discovered. Rev. Biol. Trop. 63 (Suppl. 2): 87-114. RESUMO

This paper reports on some sea cucumbers (Holothuroidea) in the collections of the NHMUK, taken off the coast of Ghana. Most result from collections around Accra during the years 1949-52. A couple of other species are included. Altogether six species are here reported, viz. Holothuria (Holothuria) dakarensis Panning, 1939; Holothuria (Semperothuria) imitans Ludwig, 1875; Holothuria (Vaneyothuria) lentiginosa lentiginosa (von Marenzeller,1893); Stereoderma congoana (Heding, 1935); Ocnus cruciformis Thandar n. sp. and Leptopentacta cabindaensis (Cherbonnier, 1949) herein transferred to a new genus Cherbocnus Thandar erected for this purpose. Another West African species, Cladodactyla monodi Cherbonnier, 1950 is referred to Stereoderma. All cucumariids are West African forms whereas the holothuriids are shared with the North-West Atlantic Ocean.

RESUMO

This is a taxonomic work on the aspidochirotid sea cucumbers from shallow waters in Northeast Brazil. We studied 165 specimens, classified into 2 families, 3 genera, 7 subgenera and 9 species. Descriptions, illustrations and literature data are included for each species, in addition to ecological notes and information on their distribution in the northeast coast of Brazil. A taxonomic key is provided for the identification of the species.
